小RNA在細菌致病機制上之作用機轉

摘要 The role of RyhB, a small non-coding RNA, in the regulation of ferric uptake 3 regulator (Fur) was investigated in Klebsiella pneumoniae strain CG43. In this study, 4 we found that expression of ryhB was directly repressed by Fur-Fe(II) in K. 5 pneumoniae. In the downstream pathways, RyhB repressed the expression of the TCA 6 cycle genes acnA, sdhA, and fum, and reduced the growth of K. pneumoniae on 7 succinate and fumarate. RyhB also regulated the resistance to hydrogen peroxide. 8 qRT-PCR showed that overexpression of RyhB activated the expression of sodA, but 9 repressed sodB, katE, and katG in K. pneumoniae. In a fur K. pneumoniae strain 10 with elevated capsular polysaccharide (CPS) production, deletion of ryhB reduced 11 CPS production. In addition, overexpression of RyhB activated orf1 and orf16 in the 12 CPS production gene cluster. Deletion of ryhB also reduced the expression of several 13 genes corresponding to three iron acquisition systems in K. pneumoniae, and resulted 14 in reduced siderophore production. This is the first study to demonstrate that RyhB 15 participates in the Fur regulon that controls TCA cycle activity, resistance to oxidative 16 stress, CPS biosynthesis, and iron homeostasis in K. pneumoniae.
